As far as I recall, you just create your playlist in iTunes (click the big + in the bottom left hand corner, I think), copy the tunes you want into it from your music library window and then, once you synch it with the iPod, you should see the playlist (under Playlists, of course) on your iPod.

You can also do it on the iPod itself, if you're out and about and nowhere near iTunes - IIRC its called an "On the Go" playlist.
Scroll to the song you want to add, then press and hold (for a few seconds) the select button in the centre of the jogwheel. Repeat for every song you want to add.
